小儿清肺合剂对幼鼠呼吸道合胞病毒炎性肺损伤中肺组织的病毒载量及炎症因子的影响

Effect of Xiaoer Qingfei mixture on the viral load and inflammatory factors in lung tissues of young rats with respiratory syncytial virus inflammatory lung injury

摘要: 目的 探讨小儿清肺合剂对幼鼠呼吸道合胞病毒(RSV)炎性肺损伤中肺组织的病毒载量及炎症因子的影响。方法 选取18只幼龄SD大鼠随机分为3组,正常组、RSV组、小儿清肺合剂干预组,每组6只。采用滴鼻法建立RSV炎性肺损伤模型;采用实时荧光定量聚合酶链式反应(PCR)检测大鼠肺组织中病毒载量;苏木精-伊红染色法观察肺组织病理改变;酶联免疫吸附法检测肺泡灌洗液中白细胞介素-8(IL-8)、肿瘤坏死因子α(TNF-α)含量,观察小儿清肺合剂对上述指标的影响。结果 (1)病毒载量方面,与正常组比较,RSV组、小儿清肺合剂干预组肺组织中RSV含量均明显增加,差异有统计学意义(P<0.05);与RSV组比较,小儿清肺合剂干预组肺组织中RSV含量明显降低,差异有统计学意义(P<0.05)。(2)病理损伤及湿干比方面:与正常组比较,RSV组、小儿清肺合剂干预组病理损伤评分及肺湿干比均增高,差异有统计学意义(P<0.05);与RSV组比较,小儿清肺合剂干预组损伤明显改善,差异有统计学意义(P<0.05)。(3)肺泡灌洗液中IL-8、TNF-α的含量方面,与正常组比较,RSV组、小儿清肺合剂干预组IL-8、TNF-α含量明显升高,差异有统计学意义(P<0.05);与RSV组比较,小儿清肺合剂干预组IL-8、TNF-α含量明显降低,差异有统计学意义(P<0.05)。结论 鼻腔滴鼻RSV可成功建立幼鼠RSV炎性肺损伤模型;小儿清肺合剂干预后可降低幼鼠RSV炎性肺损伤中肺组织的病毒载量,抑制炎症因子水平,改善肺损伤程度。

Abstract: ObjectiveTo investigate the effect of Xiaoer Qingfei mixture on viral load and inflammatory factors in lung tissues of young rats with respiratory syncytial virus(RSV) induced inflammatory lung injury.MethodsEighteen young SD rats were randomly divided into three groups:normal group,RSV group,and the intervention group of Xiaoer Qingfei mixture,with 6 rats in each group.RSV inflammatory lung injury model was established by nasal drip method. Real-time PCR was used to detect the viral load in rat lung tissues. Hematoxylin-eosin staining(HE staining) was used to observe the pathological changes of lung tissue. ELISA method was used to detect the content of IL-8 and TNF-α in BALF. The effect of Xiaoer Qingfei mixture on the above indexes was observed.Results(1)In terms of viral load: compared with the normal group,the RSV content in the lung tissue of the RSV group and the intervention group of Xiaoer Qingfei mixture was significantly increased(P<0.05);compared with the RSV group,the RSV content in the lung tissue of the intervention group was significantly lower(P<0.05).(2)In terms of pathological injury and wet-dry ratio:compared with the normal group,the pathological injury score and the wet-dry ratio of the lung in the RSV group and the Xiaoer Qingfei mixture intervention group were higher(P<0.05);compared with the RSV group,the injury in the intervention group was significantly improved(P<0.05).(3)In terms of the content of IL-8 and TNF-α in BALF: compared with the normal group,the content of IL-8 and TNF-α in RSV group and the intervention group of Xiaoer Qingfei mixture was significantly higher(P<0.05);compared with RSV group,the content of IL-8 and TNF-α in the intervention group of Xiaoer Qingfei mixture was significantly lower(P<0.05).ConclusionRSV intranasal drip can successfully establish RSV inflammatory lung injury model in young rats. The intervention of Xiaoer Qingfei mixture can reduce the viral load of lung tissue in RSV inflammatory lung injury of young rats, inhibit the level of inflammatory factors, and improve the degree of lung injury.
