盛丽先经验方清消Ⅰ号对肥胖小鼠血脂及炎症因子的影响

Effect of Sheng Lixians empirical formula Qingxiao on blood lipids and inflammatory factors in obese mice

摘要: 目的 研究名中医盛丽先教授经验方清消Ⅰ号对高脂饮食诱导的肥胖小鼠血脂及炎症因子的影响。方法 将32只C57BL/6J小鼠,随机分为对照组,模型组,中药低、高剂量组4组,每组8只。模型组给予高脂饮食8周,建立肥胖小鼠模型。于第9周起给药干预,对照组、模型组给予等剂量生理盐水灌胃,中药低、高剂量组分别灌胃5.04、10.08 mg/(g·d)清消Ⅰ号水煎剂,共干预8周。16周末,各组小鼠同时测定Lee′s指数,分离腹腔脂肪,苏木精-伊红染色观察病理。生化法测定血清胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白的含量,酶联免疫吸附法(ELISA)测定血清中肿瘤坏死因子α(TNF-α)和白细胞介素-6(IL-6)的表达水平。结果 模型组小鼠的体质量、肝脏湿质量、内脏脂肪质量以及Lee′s指数与对照组相比,均有显著增加;血清胆固醇、甘油三酯和低密度脂蛋白含量显著增高,高密度脂蛋白含量显著降低,血清TNF-α及IL-6水平升高(P<0.05)。经清消Ⅰ号治疗后,与模型组比较,小鼠体质量、肝脏湿质量、内脏脂肪质量以及Lee′s指数均明显减低;血清胆固醇、甘油三酯和低密度脂蛋白含量明显降低,高密度脂蛋白含量升高。血清TNF-α及IL-6水平降低(P<0.05)。结论 清消Ⅰ号对高脂饮食引起的肥胖小鼠有较好的治疗作用,可能是通过降低血清炎症因子TNF-α及IL-6的表达水平,调节血脂,从而改善肝脏病理,减轻体质量。

Abstract: ObjectiveTo study the effect of Professor Sheng Lixian′s empirical formula Qingxiao on blood lipids and inflammatory factors and the related mechanisms in an obese mouse model induced by a high-fat diet.MethodsA total of 32 C57BL/6J mice were randomly divided into a control group,a model group,and a low-dose and a high-dose group of Qingxiao ,with 8 mice in each group.The model group was given a high-fat diet for 8 weeks to establish an obese mouse model.Starting from the 9th week,medication intervention was administered.The control group and model group were given equal doses of normal saline by gavage,while the low-dose and high-dose groups of Qingxiao were given 5.04 mg/g·d and 10.08 mg/g·d Qingxiao decoction by gavage,respectively.The intervention lasted for a total of 8 weeks.At the end of the 16th week,the Lees index was measured simultaneously in each group of mice,abdominal fat was separated,and HE staining was performed to observe the pathology.Biochemical method was used to measure the levels of serum CHOL,TG,HDL-C,LDL-C,and ELISA method was used to detect serum TNF-α and IL-6 levels.ResultsCompared with the control group,the model group mice showed significant increasein body mass,liver wet mass,Lee′s index,and visceral fat mass;serum CHOL,TG,and LDL-C levels were significantly increased,while HDL-C levels were significantly reduced;the levels of TNF-α and IL-6 increased.Compared with the model group,the body mass,liver wet mass,Lees index,and visceral fat mass of mice after the treatment with Qingxiao significantly decreased;serum CHOL,TG,and LDL-C levels significantly decreased,while HDL-C levels significantly increased;TNF-α and IL-6 levels decreased(P<0.05).ConclusionQingxiao can reduce serum TNF-α and IL-6 levels and regulate blood lipids,improve liver pathology,and reduce body mass,and thus have a good therapeutic effect on obese mice caused by a high-fat diet.
